No, you do not have to use all 4 slots in your iMac. However, I haven't personally seen a 16GB RAM chip used in your model iMac and I've seen a lot of iMacs in my time, so others here may have to comment on whether they've actually tried it. Typically, to get to the maximum 32GB RAM, users have 4 x 8GB RAM chips installed (one in each slot). I have a late 2013 iMac with 16gb ram and upgraded 2.9 i5 processor that had a fusion drive. I added a new 2tb ssd last month. I also added a 500gb nvme ssd (or whatever the MacBook Pro equivalent drive is called) that I had spare from my late 2013 MacBook Pro as my iMac version has a slot for the nvme type ssd drive on the logic board.
